Watch Supernatural S6E3: The Third Man
Sam and Dean goes calling Castiel for help after a case they are investigating, that involves the death of some police officers, seems to have been caused by the plagues that resembles the ones from Egypt. Castiel then confirms their suspicions and tells them that Heaven is now in a state of chaos and that God's weapons have been stolen and one of it is the Staff of Moses which have been used to kill the cops. Now, as they try to recover the Staff, they again comes face to face with an old nemesis.
